簸箕柳与杞柳种间杂交可行性分析毕业论文

摘 要

簸箕柳(Salix suchowensis) 与杞柳(Salix integra)都为杨柳科,柳属(Salix),属于我国重要的经济、生态树种。杞柳主要分布于我国河北燕山,黑龙江、辽宁、吉林三个省份;簸箕柳主要分布在江苏、山东、浙江、河南等省。簸箕柳和杞柳皆是1年生便能开始开花结实,是为林木遗传研究的优选植物。论文分析植株的形态学差异,亲本与杂交成功的子代的对比,并且利用Indel标记染色体的位点、PCR扩增等位基因、利用6%聚丙烯酰胺变性胶进行电泳检测以及SSR引物筛选对簸箕柳与杞柳种间杂交的可行性进行分析。论文选取的144对Indel标记全都匹配成功,根据SSR看出看出父本基因型AA,母本基因型BB,F1代位点为AB型或者是父本基因型为AB, 母本基因型CD,F1代位点为AC、AB、BC、BD型。以父本为杞柳,以母本为簸箕柳的种间杂交组合可以进行,种间杂交的亲和性高,后代种子的存活率高;相反,以母本为杞柳,父本为簸箕柳的种间杂交组合很难进行,种间杂交的亲和性低,后代种子的存活率低。论文对不同杂交方式对种间杂交效果的影响进行了讨论。

Feasibility analysis of Interspecific hybridization between Salix suchowensis and Salix integra

ABSTRACT

Both Salix suchowensis and Salix integra belong to Salicaceae,Salix. Belonging to one of the important economic and ecological tree species in China. Salix integra mainly distributed in China's Hebei Yanshan and Heilongjiang, Liaoning, Jilin these three provinces. Salix suchowensis is mainly distributed in Jiangsu, Shandong, Zhejiang, Henan and other provinces. Salix suchowensis and Salix integra both can start flowering and fruiting when they are one-year-old, is an excellent plant for providing research on forest genetics.

In this paper, the morphological differences of the plants were analyzed, and the parents were compared with the successful progeny of the hybrids.The feasibility of hybridization between Dustpan and Salix was analyzed by using Ctab method to extract genomic DNA, sites of indel labeled chromosomes, PCR amplification allele, electrophoresis detection using 6% polyacrylamide gel and SSR primer screening.
The main result was that the selected 144 pairs of Indel markers all matched successfully, according to the SSR screening pictures, the male parent genotype AA, the female parent genotype bb,f1 is AB type or the male parent genotype is AB, the female parent genotype cd,f1 is AC, AB, BC, BD type.On the whole, with the success of the hybrid plant pictures, with the male parent as the Qi Liu, the female parent as a dustpan of the interspecific hybrid combination can be carried out, the affinity of interspecific hybridization, the survival rate of progeny seed is high, the isolation mechanism of small species;On the contrary, with the female parent as the Qi Liu, the male parent is the interspecific hybridization combination of dustpan, the affinity of interspecific hybridization is low, the survival rate of progeny seed is low, and the isolation mechanism between species is large.
